Optimal Concentration of AAGP™ Required for Increased Survival of Keratinocytes Under Normal Growth Conditions.
Anti Aging Glycoproteins (AAGPs™) are small, stable synthetic replicas of AFGP (antifreeze glycoproteins), which have been found to have cell protective properties in nature. With this in mind, our aim was to investigate the affect of a range of concentrations of AAGP™ on keratinocyte cell numbers under normal growth conditions.
Keratinocytes were seeded at a concentration of 25,000 cells/well (day -3) and incubated with 0, 1, 2, 5, 10 or 15mg/ml of AAGP™ (day 0). The effect of AAGP™ on the rate of cell proliferation was measured using the Trptan Blue exclusion method at 24 and 48 hours post exposure. Three wells were counted per concentration and 3 counts were performed per well.
In keratinocytes exposed to 1 and 2 mg/ml AAGP™ there was a significant (28% and 50% respectively) increase in cell number, in comparison to the control (0mg/ml AAGP™), 48 hours post exposure.
This would suggest that exposure of keratinocytes to 1 or 2mg/ml AAGP™ either increases the rate of cell proliferation or protects against normal cell death. However, previous experiments have shown AAGP™ does not affect cell doubling time. Thus, these results together suggest AAGP™ gives a cell survival advantage in keratinocytes - under normal growth conditions.
